Whole transcriptome sequencing of 20 paried involved/uninvolved psoriatic         and 20 normal control skin tisues.We focused on the relationship between gene expression and DNA methylation of 20PP/PN and 20UC, all of  which have been evaluated by DNA methlation array and trancriptome sequencing. In this study we did not analyze the sequence change like gene mutation, fragement variation, gene fusion and alterative splicing, even though RNA sequencing data contains these info.  Library construction:  Total RNA was isolated from the 20 PP / PN and 20 NN skin samples using the RNAeasy Mini Kit (Qiagen, Chatsworth, CA, USA) according to the manufacturer’s instructions. The RNA concentration and purity were measured using a NanoDrop ND-1000 spectrophotometer (Thermo). The RNA integrity was assessed according to the intensity and shape of the 28S and 18S rRNA bands by agarose gel electrophoresis and analysis on the Bioanalyzer 2100 (Agilent Technologies). The RNA samples with an integrity number (RIN) larger than 6.0 were used for transcriptome sequencing. The cDNA libraries were constructed using the TruSeq library generation kits following the manufacturer’s instructions (Illumina, San Diego, CA). Several libraries were pooled, and then bridge amplification was performed using cBot (Illumina, Inc, San Diego, CA) according to the manufacturer's standard cluster generation protocols. After hybridization of the sequencing primers, base incorporation was carried out on a HiSeq 2500 Sequencer (Illumina, Inc, San Diego, CA) following the manufacturer's standard sequencing protocols (101 cycles of sequencing per read generating paired-end reads that included 100 bps at each end and 6 bps of the index tag).
