Background. Nuclear factor I-A (NFI-A), a phylogenetically conserved transcription/replication protein, plays a crucial role in mouse brain development. Previous studies showed that disruption of the Nfia gene in mice leads to perinatal lethality, corpus callosum agenesis, and hydrocephalus.

BAT is not thermogenic under an unstimulated state; whereas can be quickly activated to generate heat after sympathetic activation. CL316,243 is a specific agonist of beta3 adrenergic receptor on brown adipocytes. Treatment of CL316,243 in mice leads to potent and specific activation of BAT. The goal of this project is to characterize acute gene expression alterations in the early stage of BAT thermogenic activation.

View SamplesDuring embryogenesis the heart forms as a linear tube that then undergoes multiple simultaneous morphogenetic events to obtain its mature shape. To understand the gene regulatory networks (GRNs) driving this phase of heart development, during which many congenital heart disease malformations likely arise, we conducted an RNA-seq timecourse in zebrafish from 30 hpf to 72 hpf and identified 5861 genes with altered expression. We clustered the genes by temporal expression pattern, identified transcription factor binding motifs enriched in each cluster, and generated a model GRN for the major gene batteries in heart morphogenesis.

Premyelinating oligodendrocytes were acutely purified by immunopanning approach from P12 Olig2-Cre; TFEB F/+ or P12 Olig2-Cre; TFEB F/F mouse brains. Bulk sequencing were performed with 4 biological replicates per genotype.
